Interleukin 2 (IL-2) and interleukin 12 (IL-12) are rendered membrane-bound (mb) by fusion to the transmembrane domain of the transferrin receptor with an interceding (G4S)3 linker. The mRNAs are delivered using a microfluidic membrane deformation technique that temporarily disrupts the cell membrane. Each mRNA comprises a 5'-cap, a synthetic 5' untranslated region containing a Kozak sequence, a coding region and a 3' untranslated region (UTR) derived from the murine alpha globin UTR. The cell population phenotype is CD45+, comprising primarily T lymphocytes (CD3+), monocytes (CD14+), natural killer (NK) cells (CD56+), and B lymphocytes (CD19+)
