THE CELLS ARE MECHANICALLY AND ENZYMATICALLY ISOLATED FROM RESECTED TUMOUR BIOPSY OF PATIENTS WITH METASTATIC MELANOMA AND CULTURED IN MEDIA CONTAINING RECOMBINANT INTERLEUKIN 2 (IL-2) AND FOETAL BOVINE SERUM (FBS). THE CELLS ARE ACTIVATED USING AN ANTI-CD3 ANTIBODY AND IRRADIATED ALLOGENEIC PERIPHERAL-BLOOD MONONUCLEAR CELLS (PBMCS) IN MEDIA SUPPLEMENTED WITH HUMAN AB DONOR DERIVED SERUM AND IL-2. THE T LYMPHOCYTES (>80%) ARE A HETEROGENEOUS MIXTURE OF CD4+ AND CD8+ TUMOUR-INFILTRATING LYMPHOCYTES (TIL) PREDOMINANTLY WITH EFFECTOR MEMORY AND CENTRAL MEMORY CELL SUBSETS. THE T LYMPHOCYTES DEMONSTRATE THE ABILITY TO KILL AND IMPEDE TUMOUR GROWTH IN A CO-CULTURE BASED CYTOTOXICITY ASSAY
